• Investigate oncology epigenetic targets, design and execute in vitro cell-based assays, validate targets and investigate the mechanism of action • Lead projects, establish methods to in vitro induce T cell exhaustion, screen and identify compounds that can reinvigorate exhausted T cells. • Optimize CRISPR-CAS9 gene editing in cell line and primary cells to validate epigenetic targets • Design and execute in vivo mouse models to explore applications of compounds combination with checkpoint inhibitors in immunotherapy; investigate T cell exhaustion, Treg, DC, and myeloid cells in tumor environment, and the impacts of compounds on tumor infiltrating lymphocytes and therapeutic mechanisms.

• Design and execute in vitro cell-based assays to validate targets and investigate the mechanism of action. • Investigate T cell exhaustion, macrophage and myeloid derived suppressor cells (MDSC) in tumor environment. • Establish and optimize in vitro high throughput methods to screen compounds. • Support in vitro functional genomics screening programs.

• Design and execute in vitro assays and in vivo murine models to investigate therapeutic mechanisms of engineered immune cells. • Engineer B cell to a potent antigen-presenting cell in tumor immunotherapy and preventative vaccine. • Develop new methods to improve dendritic cell based immunotherapy. • Develop methods, including in vitro, in vivo and ex vivo, to evaluate the applications of intracellular delivery methods in tumor immunotherapy. • Manage outside projects with CROs.

• Study on antiviral functions of mucosal innate immunity, and how we may harness these potent antiviral properties in the context of a preventative vaccine. • Investigate the mechanisms of natural killer cell memory in non-human primates. • Investigate pDCs functions in specific anatomic tissue microenvironments.

Study on antiviral functions of innate immunity and mucosal immunity in the gastrointestinal tract, using rhesus macaques as models. Identify subsets of innate lymphocyte cells (ILCs) in the gastrointestinal tract of rhesus macaques. Dendritic cells and HIV/SIV infection.

B cell and antibody studies in HIV vaccine. Established B cell Elispot to detect HIV gp120 specific memory B cells and plasma cells using mice model. Identify and isolation of antigen-specific B cell using flow cytometry. Analyze the quantity and quality of HIV gp120 specific B cell and antibody response.

Managed patients infected with various pathogens, including HIV-1, HCV, HBV, SARS, EV-71, et al. Involved in international clinical trials as a sub-investigator (TMC278-TiDP6-C209; TMC278-TiDP6-C215). Acute HIV infected patients-MSM cohort study. Research on T cell function and response during HIV infection.
