Project: Molecular and Dynamic Proteomics by High-throughput Techniques • Generated and analyzed genome-wide protein interactome and gene regulatory networks by high-throughput yeast two-hybrid system (Y2H), LUMIER, mass spectrometry assays, ELISA, Protein-fragment Complementation Assays (PCA), and CRISPR library screening. • Generated SARS-CoV-2-human protein-protein interactome network (published in Nature methods, and submitted to Nature Biotechnology) • Screened mutational hotspots and integrated with computational approaches to systematically affect protein-protein interactions in cancer (published in Genome Research and Nature Genetic). Design and troubleshoot molecular and cellular assays for screen experiments. • Generated stable expression cell library using CRISPR system for studying cancer driving genes • Utilized Tecan Freedom Evo bio-robot to perform 96-well plate-based high-throughput DNA extraction, Y2H, Plate-seq and Next Generation Sequencing Library preparation • Validated novel protein-protein interactions using high-throughput in cellulo fluorescence-based PCA assay and in vitro luminescence-based assay and Co-IP assay • Experienced in molecular biological and recombinant DNA techniques including restriction enzyme digestion, cloning, DNA preparation, PCR, sequence analysis, expression of recombinant proteins in bacteria, purification of proteins
