About

I am a multidisciplinary researcher with over 20 years of professional experience in various biology subjects, covering from molecular/cell biology to structural biology. I am currently working on Class III PI3K complexes (VPS34 complexes) using cell biological, biochemical, biophysical, and structural biological methods in the laboratory of Roger Williams, FRS at the MRC-LMB, Cambridge, UK. I usually work as a team member, whereas I can also work independently when necessary. I am also on the Green Committee at the MRC-LMB, where I am keen on laboratory sustainability. Main accomplishments: -Developed an assay method to quantitatively measure lipid kinase activities (Ohashi et al., eLife, 2020) -Developed protein expression and landmarking methods for cryo-EM using a thermotolerant yeast, K. marxianus (Baretić et al., Nat Commun, 2016) -Developed a multiple-protein expression system using S. cerevisiae (Rostislavleva et al., 2015, Science) -Characterised membrane traffic pathways in autophagy (Ohashi and Munro, MBC, 2010) -Developed transgenic plants that can inducibly generate root hairs at any organs (Ohashi et al., Science, 2003) Main skills: -Problem solving -Supervision -4000+ DNA constructs for cell biology, structural biology, and transgenic organisms etc.. -Developing protein expression systems using eukaryotic cells (mainly yeast and mammalian cells) -Protein purification optimisation for large protein complexes with multiple subunits ( ~ 1.5 MDa) -Nanobody characterisation (screening, engineering etc...) -Cell biology skills -Molecular biology skills -Biochemistry skills -Yeast and plant genetics Other profiles: -ORCID: https://orcid.org/my-orcid?orcid=0000-0002-2288-130X -Research Gate: https://www.researchgate.net/profile/Yohei-Ohashi-3 -Loop: https://loop.frontiersin.org/people/319669/overview

MRC Laboratory of Molecular Biology

Laboratory of Roger Williams -Developed multiple protein expression systems using S. cerevisiae for studying Class III PI3Ks, along with protein purification optimisation and cell biology (Rostislavleva et al., 2015, Science) -Developed a new protein expression system using a thermotolerant yeast, K. marxianus. Also developed an electron density marking method using tandem-RFP for cryo-electron microscopy (Baretić et al., 2016, Nature Comm).
