Ginkgo Bioworks, HTS, cell-based assay development • Technique guidance and support for multiple programs including: Saponin as vaccine adjuvant; Gut Microbiome and Autoimmune Disease Treatment, circular RNA for antibody and protein expression, et al. m • Established multiple assays for inflammasome pathway, Pattern Recognition Receptors (PRRs) pathways, cytokine/chemokine and other biomarkers measurement, immune function testing (T cell function testing, NK cell function testing, mouse and human antigen presentation assays, et al.), RNA and lipid nano particle immunogenicity and toxicity testing.

• Key contributor for multiple programs, including interferon pathway related programs, tumor specific NK cell killing program, tumor growth inhibition programs, and autoimmune disease programs. • Developed more than ten cell-based assays for target engagement, MOA study and compound screening. • Established multiple cytokine/chemokine quantification assays to support In Vivo PD marker quantification for eight programs. • Parts of work have been published in Science and other journals (Science. 2020:369(6506), SLAS Discov. 2021, Jun 16, Online ahead of print).

• Contributed to interferon pathway agonists and antagonists program, including STING agonist, Rig-1 agonist, MDA 5 antagonist. PCC of STING and Rig-1 agonists are on clinical trial now. • Contributed to asthma/diabetes lead identification project by developing cell-based functional assays and PI3 kinase isoform selective assays for compound screening. • Contributed to hypertension lead optimization project by exploring signaling pathways affected by lead compounds. • Assessed off-target effects of lead and clinical compounds in cell-based assays.

• Contributed to dendritic cell-based vaccine on PML treatment program. • Monitored the changes of viral load, cellular and humoral immune response in MS patients with natalizumab (Chen et al 2009 New England J Medicine). • Measured BKV specific cellular immune response in kidney transplant recipients with polyomavirus nephropathy. Successfully discovered several BKV specific CTL epitopes for BKV VP1 and large T antigen (Chen et al 2008, Lima et al 2007, Chen et al 2006). • Characterized a BKV strain which causes Capillary Leak Syndrome (Chen et al 2004, Barouch et al 2002).
