My first gene cloning experience was with PSA (https://www.sciencedirect.com/science/article/abs/pii/S0022534717397975). We cloned it using an antibody in just a few days. Because it seemed so easy, some colleagues thought it wasn’t important—until another group published it in BBRC. That taught me an important lesson: ease doesn’t equal insignificance. My Ph.D. focused on Testicular Feminization (TFM), known in humans as Androgen Insensitivity Syndrome (AIS). These patients are genetically male (XY) but develop as females due to a defective androgen receptor. Incredibly, sometimes this is caused by a single nucleotide change out of 6 billion codes. It still amazes me how such a small difference in DNA can have such a profound effect. We are probably still underestimating the power of our DNA and its influence on our health.
