**YOLT-204 (In Vivo Gene Editing for β-Thalassemia)** As a core team member, I drove assay development and lead optimization to enable precise in vivo gene editing, directly supporting preclinical-to-clinical translation. Designed and executed critical assays to evaluate editing efficiency, safety, and pharmacodynamics in preclinical models. Played a pivotal role in analyzing clinical samples to assess therapeutic impact and guide program decisions. **YOLT-401 (In Vivo CAR-T Platform)** Led in vivo CAR-T program from early discovery through candidate selection. Spearheaded the development of robust analytical assays (e.g., CAR expression, immune cell profiling) and optimized LNP delivery systems to enhance targeting and potency. Collaborated cross-functionally to screen lead candidates, establishing key proof-of-concept data to advance the platform.

• Utilized mouse model, spectual flow cytometry and statistical modeling to investigate precursor number of HSPC in native hematopoiesis and a mouse model of Fanconi Anemia. • In collaboration with Peranteau lab, generated high-titer mouse Fancc-expressing lentivirus to test fetal lentivirus injection as a therapy in a mouse model of Fanconi Anemia.
