lab and office operations

I am the lab manager of Infectious Disease laboratory, the point of contact between external contractors or internal support teams and the research team. Manage all the non-science matters of the lab: such as lab organization, biological sample management, procurement and supply chain execution, capital equipment life cycle management, lab maintenance and safety, waste stream, lab safety SOP establishment, 24/7 emergence response contact, new hire orientation and employee exit management, and so on. I am also serving as Alternate Radiation Safety Officer to oversee radiation safety program.

Antiviral Drug Discovery against Hepatitis B virus, Coronavirus: o Developed transfection assay, ELISA assay, qRT-PCR assay, cytopathic effect assay for antiviral quantification. o Performed compound screening and MOA studies, advanced the projects to clinical trials. o Setup external contract for SARS-Cov-2 projects and managed the project. Key person for company initial lab space planning and lab setup. o Managed the consumable stockroom.

Drug discovery against Influenza virus, RSV, Coronavirus, Dengue virus, and Chikungunya virus: o Supported ongoing RSV and influenza projects by performing regular replicon assays, neuraminidase assay, cytopathic effect assay. o Developed Assays for dengue virus quantification through qRT-PCR. o Compound resistance profiling

Antiviral Drug Discovery against HBV: o Developed assay protocols for HBV drug discovery: Assay development for HBV HBsAg, HBeAg, intracellular and extracellular HBV DNA and RNA quantification, cccDNA quantification. o Performed HTS compounds screening and lead identification. o Supported the Clinical trial of first in class HBV capsid inhibitor which lead to acquisition of Novira by Janssen. Key person for company initial lab space planning and lab setup. Managed the consumable stockroom.

Responsibility: Anti-viral drug discovery. Achievements: • Developed ChemiLuminescence ImmunoAssay (CLIA, ELISA) and AlphaLISA assay for extracellular viral protein detection and quantification, Immunofluorescence Assay for intracellular viral protein detection, Quantigene assay and Taqman Assay for RNA and DNA quantification, viewRNA assay for in-site RNA hybridization, CPE-based assay and Renilla Luciferase Assay for antiviral inhibition; Developed and optimized the cell-based antiviral assays for high-throughput screening and medium-throughput drug screening. • Constructed mutant recombinant HBV-Baculovirus and characterized this non-infectious recombinant virus in primary hepatocytes, leading to successful completion of high-throughput screening of millions compounds. • Accomplished thousands of compound screening against different virus (HBV, Dengue, Influenza) in cell-based assays, identified one candidate leading to Dengue virus clinical trial, and dozens of lead hits for HBV.HBsAg inhibition. • Established RNA quantification assays to validate the RNA inhibition effect for HBV siRNA projects, and screened over 50 siRNAs against HBV RNAs, leading to one clinical candidate ARC-520. • Programmed all Hamilton/Biomek methods for liquid handling, expedited the screening process by 6x times.

• Developed Medium throughput antiviral screening assays: Quantitative assays for HBV HBsAg/HBeAg, and RNA/DNA. • Established different Assay systems for HBV drug discovery: Baculovirus-HBV recombinent system and HBV stable cell line based system. • Screened thousands of compounds for HBV inhibitors. • Programmed all the Hamilton methods for screening.

Anti-viral Drug Discovery: • Developed Medium throughput antiviral screening assays for Dengue virus, Influenza virus, and HBV: CPE-based assay, ELISA, AlphaLISA, bDNA assay, etc. • Established different Assay systems for HBV drug discovery: Baculovirus-HBV recombinent system and HBV stable cell line based system. • Screened thousands of compounds for HBV and Dengue virus inhibitors. • MOA study of potent Dengue or HBV inhibitors.

• Characterizied RNA cap methylation of West Nile virus: Constructed over 20 West Nile Virus Methyltransferase mutants, performed protein expression and purification, analyzed all mutant proteins with biochemical enzyme assay, characterized RNA cap methylation of West Nile virus Methyltransferase and demonstrated the substrate re-position mechanism. • Crystallization of West Nile Virus methyltransferase. • Rescue of Reassortants from a Highly Pathogenic H5 flu Virus. • Published 4 papers in Virology and Journal of Virology.
