During my PhD I worked on the structural characterization of a 2.2 MDa ion channel with small molecule modulators as well as antigen-antibody complexes for SARAS-CoV-2. Through this work I purified ion channels directly from native tissue and structurally characterized the interactions and effects of small molecule modulator binding using cryo-EM. Further, I leveraged X-ray crystallography for characterizing ligand interactions with truncated protein domains and used these structural results for the optimization of small molecules. Through completing this degree I developed skills in: -Protein Expression (bacterial, Expi293) -Protein purification (from native tissue, various affinity tags, IEX, SEC) -Biological assay development for in vitro and cellular measurements -Structural characterization (Cryo-EM and X-ray crystallography)
