I am currently a scientist at Benitec Biopharma, working on manufacturing and process development on AAV-based gene therapy. Before I joined Benitec Biopharma. I complete PhD degree in university of Arkansas where I have been working on the fouling detection, modeling, visualization during tangential flow filtration and alternating tangential flow filtration. Before joining University of Arkansas, my research is foam fractionation, which is a novel method for protein enrichment. Cell harvesting the beginning of the downstream purification process for most of protein-based pharmaceutics. This process is more challenging because of the increase of cell density and cell culture duration, especially with the adaption of alternating tangential flow filtration(ATF) technique. The most limiting factor of this process is the fouling of the membrane. This work tries to answer 2 main questions: what is the foulants and how they fouled the membrane. Experimental works include the 2D SDS-PAGE and foulants identification by LC-MS. With a better understanding of the first question, the pharmaceutical manufacturer could eliminate foulant that is identified from this work. The second question could help the membrane manufacturer to design a better membrane with higher mass transfer.
