Listeria-based vaccines are highly promising vectors for cancer immunotherapy. The goal of this NIH-funded project was to develop a novel Listeria-based vaccine targeting CD105 to treat Renal cell carcinoma (RCC). CD105 is strongly expressed on proliferating endothelial cells such as those in tumor vasculature but, also on RCC tumor cells themselves. Therefore, targeting CD105 is a promising strategy for RCC cancer treatment. Our Listeria-based CD105 vaccines infect dendritic cells (DCs) that will process and present CD105 peptides on MHC class I and II molecules. These mature DCs will then activate CD105-specific CTLs that target both tumor cells and tumor-associated vasculature on RCC tumor models. My responsibilities • Culture primary immune cells (T cells and dendritic cells), tumor cells lines (human and murine), and bacteria (E. coli and L. monocytogenes). • RCC tumor models (subcutaneous and orthotropic) • Flow cytometry including staining cells, operating BD Fortessa, developing compensation panels, and data analysis • Design and conduct immune-related experiments, like ELISA (serum cytokines), IHC, Western blot, etc. • Manage large animal colony including knockout and transgenic mice and collect tissues from animals to perform genotyping (ear punches, blood samples, tail snips) • DNA and RNA purification for transfection, sequencing, and quantitative PCR • Conduct animal experiments under IACUC-approved protocols using Listeria-based vaccines, antibody-based receptor blockade, etc. • CD8+ T cells, CD4+ T cells, naïve CD4+ T cells sorting by magnetic-bead separation • Train new students 项目1介绍:肾细胞癌(RCC)是美国肾癌最常见的种类,发病率并逐年攀升。CD105是一种特异分子大量表达于RCC的癌细胞和癌组织附近脉管系统中,李斯特菌疫苗作为一种优秀的载体可以特异感染树突状细胞并诱导产生对CD105特异的CD4和CD8阳性T细胞,从而双重追踪和杀灭癌细胞和肿瘤相关血管上皮细胞。 工作职责:1通过体外细胞实验探索感染过程的细胞信号通路以及相关细胞因子的参与过程;2通过动物实验验证构建的减毒李斯特菌疫苗的活性和毒性;3通过免疫类试验检测细胞因子在感染过程的差异;4实验室管理,试剂购买以及实验室统筹技术和质量控制。

Adoptive cell transfer (ACT) is the transfer of cells into a patient to treat disease. The cells used in ACT originate either from the patient or another individual. The cells are most commonly derived from the immune system with the goal of improving immune functionality and characteristics. In autologous cancer immunotherapy, T cells are extracted from the patient, genetically modified to target the cancer cells, cultured in vitro, and returned to the same patient.
