Demonstrated the importance of highly conserved C-terminal tetrapeptide to functional enzymatic tertiary structure via genetic manipulation approach. Developed and optimized a purification protocol for a native protein with FPLC-automated column chromatography, including hydrophobic, ion-exchange, size-exclusion and affinity columns. Characterized the purified protein with a variety of technologies, including SDS-PAGE, native-PAGE, Western blot, in gel activity assay, enzyme kinetic assay, substrate specificity, inhibitor regulation, Km and Ki values determination. Resulted three (3) first-authored peer-reviewed scientific publications.
