1) Design, organize, execute, and interpret the chemistry involved in the synthesis of novel backbone-modified oligonucleotide-peptide conjugates. Solid phase synthesis (phosphoramidite method) and purification procedures (including prep HPLC) and analytical methods (HPLC; LC/MS; MALDI; quantitative UV/Vis; and 1H, 13C, 31P NMR) for phosphorus-modified and for fluorescein-labeled oligonucleotides. Grow and maintain mammalian cell cultures and carry out cell transfection experiments, quantification of cell uptake by FACS, fluorescence microscopy. 2) Design, synthesis and characterization of novel phosphoramidite monomers for the optimization of long DNA sequences.
