Main work：Generation of functional antibodies targeting GPCR • Planned specific projects in the area of antibody libraries construction, selection, characterization, expression and purifications. • Explored several noval screening technologies that are suitable for screening most antibody • Constructed full length antibody and characterized these antibody by ELISA and FACS,etc • Purified proteins and prepared the antigen protein (from E.coli and 293F/E cells) with HIS column, IgG column, SDS-PAGE,Western Blot,etc

Main work：Setup the antibody phage display technology platform • Designed and constructed antibody V-gene libraries, CDR mutagenesis libraries • Antibody screening and affinity-maturation with phage display; high throughput screening with fluorescence activated cell sorting (FACS); affinity measurement with Surface Plasma Resonance (Biacore). Some of the antibodies are presenting promising leads that to develop broad auto-inflammation therapeutics • Explored several noval screening technologies that are suitable for optimizing most humanized antibody • Constructed gene expression system: including gene cloning and gene mutation, then expressing in E.coli, and 293F expression system • Purified proteins and prepared the antigen protein (from E.coli and 293F cells) with HIS column, IgG column, SDS-PAGE,Western Blot,etc

Project: Set up humanized antibody platform for tumor necrosis factor-α (TNF-α) and interleukin-12 (IL-12). Main work： • Established Fab fragment screening platform. • Constructed humanized antibody library. • Cloned dozens of expression plasmids containing antibody heavy chain • Experience in screening high affinity surrogate antibody in vitro as guidance

Project: clinical diagnosis of respiratory viruses and phylogenetic analysis. Main work: • Performed the routine diagnosis work and analysis for human respiratory viruses using high throughput molecular assays sequencing. • One significant discovery of this work was that we found a new Rhinoviruse, the related results have been published in the journal PLoSONE (July 2009).
