PhD supervised by Professor Sir John E Walker (Nobel laureate, 1997). My project was to characterise the regulation of ATP hydrolysis of the F-ATP synthase from Caldalkalibacillus thermarum. The ATP synthase from this bacteria only acts as a synthase, unlike mitochondrial F-ATP synthases. I expressed the soluble domain of the enzyme from E. coli and purified it with affinity chromatography (His-tag). I used X-ray crystallography and molecular replacement to determine the atomic structure of the bacterial enzyme in order to identify any differences between it and mitochondrial F-ATP synthase structures. Other biochemical experiments included activity assays, BCA protein quantification assays, luciferase assays, phosphate detection assay, and thermal stability assays.

Techniques: Protein expression, purification and quantification. SDS-PAGE analysis. Circular dichroism. Solid state NMR. My final year honours project involved investigating which phospholipids were part of the annular lipid shell for the potassium crystallographically-sited activation channel (KcsA).
