Education

Structural Biology
PhD supervised by Professor Sir John E Walker (Nobel laureate, 1997). My project was to characterise the regulation of ATP hydrolysis of the F-ATP synthase from Caldalkalibacillus thermarum. The ATP synthase from this bacteria only acts as a synthase, unlike mitochondrial F-ATP synthases. I expressed the soluble domain of the enzyme from E. coli and purified it with affinity chromatography (His-tag). I used X-ray crystallography and molecular replacement to determine the atomic structure of the bacterial enzyme in order to identify any differences between it and mitochondrial F-ATP synthase structures. Other biochemical experiments included activity assays, BCA protein quantification assays, luciferase assays, phosphate detection assay, and thermal stability assays.

My final year honours project involved investigating which phospholipids were part of the annular lipid shell for the potassium crystallographically-sited activation channel (KcsA).
