1. Hybridoma antibody discovery workflow. • Led three antibody discovery campaigns utilizing Hybridoma technology. • Managed the methods setting and validation of Hamilton system (Automation liquid handling system). • Performed validation of discovered antibodies using the Hamilton system, Flow cytometry, ELISA, Cytotoxicity assay, PBMC isolation, mammalian cell culture. 2. Technology Development: (accelerate antibody discovery timeline) • Optimized flow multiplexing cell binding assay for four cell lines. • Developed 384-well plate flow binding screening method and protocol for BD FACS Canto II.

[PI : Laura F. Su, M.D. Ph.D. ] Project: Reshape of the virus-specific T cell repertoire in unexposed adults by SARS-CoV-2 vaccination 1. Identified and evaluated the population of T cell repertoire with antibody staining and multicolor flow cytometry 2. Isolated over 10 PBMCs from human peripheral blood, and enriched the T cells from the PBMCs 3. Produced 4 different DR proteins alleles (MHC II) that transduce by baculoviruses and produced tetramers. 4. Troubleshot the protein production process and increased 40% of yield rate

Project : Improve iron tolerance of in-house cell line in platform medium by using adaptation strategy 1. Evaluated the cell culture performance of 3 adapted in-house cell lines in 14-day fed-batch process in Ambr ® 250 2. Verified that adapted cell line had improved cell culture performance both in platform medium and in medium with elevated iron concentration. Product quality analysis is ongoing. 3. Characterized the trajectory of adaptation and collected materials for investigating the underlying molecular mechanisms. • Tested 5 iron reagents of their impact on cell culture performance and product quality

➢ Completed two bispecific antibody related projects: 1.Development of Anti-EGFR/anti-mPEG BsAb to Actively Attract mPEGylated Nanomedicine for Synergistic Therapeutic Effect on Pancreatic Cancer. 2. The Therapeutic Effect of mCLPS1/anti-mPEG BsAb, hzmCLPS1/anti-mPEG BsAb, mCLP3S1/anti-mPEG BsAb on Triple-Negative Breast Cancer.

Project 2: The Therapeutic Effect of mCLPS1/anti-mPEG BsAb, hzmCLPS1/anti-mPEG BsAb, mCLP3S1/anti-mPEG BsAb on Triple-Negative Breast Cancer 1. Produced and verified the quality and quantity of Bispecific Antibodies (BsAb) Project 1: Development of Anti-EGFR/anti-mPEG BsAb to Actively Attract mPEGylated Nanomedicine for Synergistic Therapeutic Effect on Pancreatic Cancer 1. Constructed, qualified, and quantified the Bispecific Antibody (BsAb) 2. Tested the therapeutic efficacy of BsAb with Lipo-Dox® to human colorectal adenocarcinoma cell (HT-29) and human pancreatic cancer cell (Mia Paca-2) ➢ Used plasmid sub-cloning, vector construction, PCR, flow cytometry, ELISA, cell culture, and cytotoxicity assays to complete projects which cooperated with doctoral students and post-Docs. ➢ Cloned 5 bispecific antibodies and 2 trispecific antibodies from establishing gene map to produce the qualified antibodies for cell toxicity and treated the xenograft tumor mice model with antibodies to evaluate the efficacy of the treatments. ➢ Coordinated 6 researchers from cross-disciplinary Labs and built a strong team by communicating and dividing the work properly. ➢ Increased 20% of antibody binding affinity by practicing the codon optimization technique.

➢ Treated intensively cared patients, oral cancer patients, renal disease patients and diabetes patients through cross-functional cooperation and provided diet prescriptions ➢ Promoted health education to 60 elders in the community about how to adjust blood sugar. etc ➢ Learned to lead 15 kitchen workers in TMU hospital and optimized the cooking standard operation procedures according to HACCP guideline
