Indulged in mAb production process development: worked from cell line development, to upstream downstream processes optimization by DOE methods. Cell cell line development: either for high mAB production or cell based assays, enjoying the art of molecular biology tools for cell genetic engineering. Hand-on experience of transcriptomic data mining for identifying targets for mammalian cell productivity improvement. Upstream process development: doing medium analysis to improve medium performance and using DOE study to find optimized cell fed-batch culture conditions. Downstream process development: extensive hands-on experience using AKTA system for affinity, AEX, and CEX columns, virus filtration, Ultrafiltration/diafiltration, and DOE for key purification steps optimization. Able to work within a team and manage project that meet or exceed timeline requirement. Trained and worked with over 15 students to accomplish and design projects for 5 of my publications. Skillsets: • Molecular biology: genomic DNA isolation, quantitative PCR, vector design and construction, mutagenesis, electrophoresis, CRISPR-Cas9 based target selection and implementation • Protein biochemistry: enzyme assays, immunoprecipitation, ELISA, Western blot analysis, SDS page separation, affinity, AEX, CEX, HIC and filtration based purification • Mammalian cell culture, transfection and large-scale cell clone/pool selection experience, extensive large-scale algae culture experience • Analytical chemistry (natural products): GC-MS (for volatile chemicals), HPLC (for amino acids and mAB aggregates), Nova Flex, Vicell, Cedex, BGA and endotoxin detection

I was working with Prof. Micheal Betenbaugh in Chemical and Biomolecular Engineering Department to establish algae as a vaccine production platform and improve CHO therapeutic protein productivity by enhancing the oxidative phosphorylation pathway. Three gene targets that could improve the CHO cell productivity were identified through literature search and transcriptomic data mining. In addition,I have further improved the Algae chloroplast protein expression platform.
