Francis Eze
Graduate Research Assistant @ University of North Texas
About
I am a PhD candidate in Chemistry at UNT, specializing in medicinal chemistry and drug discovery. My current research focuses on the design, synthesis, and development of small molecules and NSGM probes targeting thrombocytopenia and thrombosis. Previously, I worked in a natural product biosynthesis lab, investigating fungal drug-resistant transporters to identify potent bioactive molecules and analyzing unusual polyketide synthases in fungi (Colletotrichum spinosum). I have proficiency in both synthetic and biosynthetic approaches to drug discovery, including genome mining, bioinformatics, isolation, screening, and characterization of small molecules and secondary metabolites. I am also proficient in analytical and molecular biology techniques, including gene cloning, heterologous expression, cell culture, bioinformatics, flash and liquid chromatography, mass spectrometry, NMR spectroscopy, surface plasmon resonance, flow cytometry, and microplate reader assays. I am passionate about translating laboratory research into therapeutics. I aim to contribute to innovative drug discovery and the development of next-generation therapeutics.

Designed and synthesized 15+ triazole-based coumarin-thiazole inhibitors targeting Factor XIIa (FXIIa), achieving nanomolar IC₅₀ values (~100% in vitro efficacy), contributing to a new lead series for contact-pathway anticoagulation. Synthesized 5 next-generation sulfated plasmin inhibitors with >300-fold selectivity for plasmin over thrombin (FIIa) and Factor Xa, a major advance toward bleeding-safe antifibrinolytic therapy. Developed high-throughput NSGM functionalization workflows enabling kinetic profiling of 20+ candidate molecules per screening cycle. Performed SAR analysis with AutoDock Vina/PyMOL docking to rationalise structure-activity trends and drive iterative library design. Applied photoaffinity labelling to map NSGM-protein binding domains and elucidate inhibitor mechanisms of action. Established LC-MS/NMR end-to-end characterisation pipeline, accelerating compound turnaround from synthesis to biological testing. Mentored 3 undergraduate researchers; set up and managed a new medicinal chemistry lab, including equipment installation, safety protocols, and team training.

Expressed and characterised nonribosomal peptide synthetases (NRPS) heterologously in Aspergillus oryzae to activate cryptic biosynthetic gene clusters. Optimised fermentation conditions for Colletotrichum spinosum CBS 515.97, leading to the discovery and structural elucidation of a novel thielavin derivative (3''-demethylthielavin) and multiple alkaloids — published in Chemistry & Biodiversity (2025). Integrated LC-MS/MS, NMR, antiSMASH, and BLAST genome mining to map gene-to-metabolite relationships in underexplored fungal systems.
