Shaw Lab, Department of Cancer Biology Generated mutants of the Insulin Receptor Substrate 1 (IRS-1) protein that is involved in breast cancer metastasis. Designed DNA primers to introduce single amino acid changes into the IRS-1 protein that will either prevent or mimic phosphorylation of specific serine residues. Generated mutants using PCR-based site-directed mutagenesis of the IRS-1 cDNA. Amplified the mutants by inoculating bacteria and purifying the plasmid DNA using standard mini-prep and maxi-prep protocols. Prepared and electrophoresed both agarose and acrylamide gels to analyze DNA and protein, respectively. Conducted protein analysis using standard western blotting protocols. Performed tissue culture using sterile technique. Completed safety training for animal and lab work.
