Main Project: Targeting hypoxia regulated sodium driven bicarbonate transporters (NDBT) to reduce breast cancer metastasis Undertook a research project into the molecular mechanisms of pH regulation, hypoxia, and cancer metastasis. The project involved investigating changes at: epigenetic, transcriptional, protein, and function levels. Fusion proteins were produced to determine binding partners assays and co-localisation. In vitro migration and invasion assays were performed; as well in vivo experimentation to determine the effect upon metastasis. Mechanistic investigations determined that NDBT knockdown or inhibition reduced mTOR activation and HIF-1 stabilization in hypoxia. This culmiated in a loss of EMT induction in knockdown cells. Furthermore, metablomics and metabolism based assays were performed to determine the effect on ATP production, lacatate, ROS levels, and mitochondrial copy number and function.
