Leveraged expertise in cell and molecular biology and bioengineered matrices through complementary in vivo and in vitro systems to understand how triple negative breast cancer metastasizes despite chemotherapy treatments. Focused on the role proteases play in altering the extracellular tumor microenvironment by digesting the extracellular matrix, other proteases, and cell surface receptors. These same proteases can also perturb the intercellular signaling and cellular organization. I found that depending on the protease, these changes can be both pro- or anti-tumorigenic. I study cells communicate intra- and intercellularly via biochemical and mechanical signals to coordinate contractility across tissues. To address these questions I have: I. Generated and screened a RNAi library of over 150 genes to find novel regulators of contractility which led to the identification and characterization of a myosin light chain kinase II. Utilized imaging, molecular, and genetic techniques to map a spatial protein oxidation gradient across the C. elegans spermathecal tissue and found a functional role for this in regulating Rho activity through oxidation of a specific cysteine III. Used extensive live imaging techniques such as time-lapse 3D microscopy and fluorescence recovery after photobleaching (FRAP), immunofluorescence of fixed samples, and generation of transgenic animals to uncover a role for filamin, an actin crosslinker, in not only regulating actin network organization, but also in organizing the localization and distribution of proteins and organelles IV. Established and managed collaborative projects within and between labs to present oral and poster presentations, and to publish several manuscripts Other duties include: Lab Manager Acted as primary liaison between sales representatives and the lab, managed additional work-study students to meet lab material and solution needs, and organized lab inventory Head of Lab Safety Managed day-to-day safety tasks like waste removal, lab cleanliness, and operations, and was responsible for formal and informal lab inspections and audits Undergraduate Mentor Mentored ten Northeastern University undergraduates and five summer researchers by developing projects and organizing experiments personalized to each student with the goal of having each student produce results that contribute to publications
