Small Molecule Drug Discovery Experience: 1) Experience in ChemPartner (Leading integrated service programs): 1a) Drived one anti-HIV program from discovery to phase II in five years. Five patent applications based on the compounds designed and prepared in ChemPartner have been published. 1b) Leading medicinal chemistry team on cancer metabolism project (ACLY). Several lead compounds have been tested in a lung cancer xenograft model and showed comparable efficacy with a clinical-in-use positive control. 1c) Leading medchem effort for the development of autophagy inhibitors as anticancer agents. Several potent compounds (in single-digit nano moles) have been demonstrated to sensitize clinical-in-use anticancer agents and compounds in late phase clinical trials with several cancer cell lines. One patent application based on our discovery has been submitted in March 2013. 2) Working Experience in Amgen: I worked in Amgen as a medicinal chemist for 12 years. I have extensive expertise in developing small molecule therapeutics, including building IP position, structure-based design, SAR study and clinical candidate selection. I am particularly effective in driving medchem programs from discovery research to clinical trial. I am creative, self-motivated with high working ethics. I am well known for solving tough problems in SAR, PK and ADMET optimization. Contributed to the developments of five clinical candidates in the major therapeutical areas, including anti-inflammatory, metabolic and anti-cancer, CNS and anti-viral. Designed and synthesized three molecules as clinical candidates. Antibody and Antibody-Drug Conjugates Discovery Experience: Target selection for antibody drug development. Phage library construction and binder selection. Human antibody generation and characterization. Target selection for antibody-drug conjugates (ADC), new linker and drug discovery, ADC generation, characterization and preclinical candidate selection.
