Development and validation of bio-analytical methods for release and stability testing of humanized monoclonal Antibodies Drug Products

My PhD Project focused on using artificial protein scaffold (AlphaRep) to mediate specific molecular recognition using directed evolution methods. I developed three main applications: - Crystallogenesis tool Building of a Protein-fragment Complementation Assay library to select quickly, in the bacterial cytoplasm, specific protein against protein targets to crystallize. Set up of a systematic approach with a protein fusion strategy. - On-Cell recognition tool Phage display selection on cells to isolate specific proteins against a protein target over-expressed on tumor cells for imaging purposes. - Intracellular recognition tool Proof of concept of the specificity for their target of those synthetic proteins inside the eukaryotic cytoplasm. Set up of culture cell in the laboratory Validation of a new tool for cellular investigation. Strong involvement in the laboratory management Team Work (technicien, engineer and assistant professor) and projects management Collaborative work with other teams useful to achieve scientific projects Teaching : Biostatistics (level1) (during three years) and Master 1 intern supervising.

Production and Purification of recombinant proteins for pharmaceutical high throughput screening (chemical compounds). - bacterial and eukaryotic expression systems - protein purification (affinity, ion exchange, size-exclusion), work with ÄKTA systems - proteins characterizations (SDS page, ELISA, DLS) Development of purification protocols for proteins of pharmaceutical interests.

Co Coordinator of the INSA Lyon iGEM first team Development of a tool to optimized purification process using synthetic biology - construction of a synthetic E.coli strain able to overproduce lipids granules exposing protein of interest.
