I am a highly skilled scientist with over ten years of expertise in cell and molecular biology, gene editing, and assay development. Currently, I am contributing my knowledge and passion to the innovative team at Turnstone Biologics, a leading biotechnology company specializing in the development of T-cell therapies for solid tumors. My specific niche is development and qualification of potency assays for our autologous cell based drug product. By doing so, I contribute directly to the advancement of our product pipeline and the delivery of life-changing solutions for patients. With a strong foundation in cell and molecular biology, my career has been a journey of scientific discovery and advancement. At Turnstone Biologics, I play a pivotal role in designing, optimizing, and validating molecular and cell-based assays. These assays are critical for characterizing the potency and safety of our groundbreaking T-cell therapies, which have the potential to revolutionize cancer treatment. My expertise extends to the field of gene editing like CRISPR. I am well-versed in mammalian cell culture and state-of-the-art confocal microscopy along with immunohistochemistry. I have a curious mind and strongly believe in team work and the art of collaboration for the success of science. Before joining Turnstone Biologics, I served as a scientific researcher at the Ottawa Hospital Research Institute. In this role, I designed and interpreted preclinical animal models of pulmonary hypertension and acute lung injury. My work in a CL-2 facility, animal handling, and functional assays have added valuable dimensions to my scientific toolkit. I hold a postdoctoral degree from IU Bloomington, where my research focused on investigating the isoform-specific role of the tumor-suppressor gene p53 in development. My dedication to scientific excellence was recognized with several honors and awards, including an honorary mention for an outstanding oral presentation and a women in science fellowship. Beyond the lab, I am passionate about science outreach and have volunteered to teach biology to high school students from lower socio-economic backgrounds. Investigated the genetic and molecular machinery underlying the isoform-specific role of the tumor-suppressor gene, p53 in development using various methods and strategies including but not limited to genetics, cellular biology, and molecular biology. • Developed assays examining gene expression by qRT-PCR, flow cytometry and FACS. • Gene editing using CRISPR/Cas9 for generating and characterizing knockouts of p53 gene. Through collaborative help from other labs, performed functional assays of these mutants leading to a manuscript currently under revision at eLife. • Determined target protein localization (immuno-labeling), ploidy (DNA labeling and flow cytometry), how many cells were actively cycling (EdU incorporation), DNA damage with and without irradiation stress (-H2Av labeling) and cell death (cleaved Caspase labeling and TUNEL), in target cell populations by flow cytometry and confocal microscopy. • Characterized functions and interactions of targeted proteins by overexpression and RNAi, in vivo and in vitro. Downstream analyses of these experiments were done through immunolabeling, SDS-PAGE, and western blotting. Analysis of sequencing was also performed in collaboration. • Peer-reviewed primary literature articles for research journals like Scientific Reports. • Performed general lab management duties like ordering supplies/ maintaining inventories and decision making on effective cost to productivity ratio especially in between research grant renewals. • Honed scientific communication and presentation skills leading to the ‘Women in Science’ fellowship and ‘Honorary Mention’ award in international and regional conferences. • Undertook mentoring and leadership roles including training of four graduate students and one research associate. • Developed active organizational skills leading to smooth operation of the departmental seminars and learned effective time-management which greatly helped in my science outreach activities.
