• Lead investigator for a therapeutic program (undisclosed indication). Independently designed and screened guides in in vitro models. Designed ELISA and JESS assays to detect therapeutically relevant protein levels in in vitro and in vivo models, alongside qPCR and NGS based assays (in collaboration with Computational team) for genotyping studies. o Completed in vivo testing at mouse and NHP level. This project involves large scale lipid-based transfection optimization in multiple cell lines for large donor template insertion such as plasmids, along with mRNA and sgRNA. Reviewing literature and incorporating new reagents to trouble shoot protocol to improve integration efficiency. o Performing ddPCR assays as one of the downstream analyses to read successful integration, other than FACS, EVOS Imaging, cytokine- expression and long-range sequencing. o As a sub-project, I am invested in developing a protocol for the synthesis of a donor template in-house (a cost-effective and efficient platform for template synthesis) • Trained in research level LNP formulations. • Experienced in managing direct report (Co-op) for more than one project. Research Associate II- Editing Technologies
• Standardized and optimized transfection procedures to multiplex edit in primary T cells. Screened numerous guides across various loci to determine the best multiplexing scenario. The success with multiplex editing impacted the team goal, thereby expanding the scope of project to identify novel proteins for precision-based gene editing approach. o Also, trained members from cross functional teams on the above platform to implement the multiplex editing strategy towards specific programs. • Implemented and evaluated reporter-based landing pad screens through extensive optimization with the goal of enabling high-through put screen for the precision-based editor platform. The reporter-based cell lines were generated using both adherent and suspension cells and screened using antibiotic selection and flow cytometry. Cell banks were eventually maintained cell banks for each line. • Involved in production of lentiviral vectors for screening novel protein for precision-based in a high-through put manner. Transduced cell lines with lentiviral vectors to generate stable cell lines with the protein of interest. • Proficient in research grade mRNA synthesis and process optimization, followed by QC and analysis using fragment analyzer. Improved the existing process of mRNA synthesis making it more time efficient. • Trained several new team members with majority of the cell biology workflow protocols, assays, and instrumentations in CRISPR Tx lab. • Presented research and development findings in quarterly portfolio reviews, exploratory science forums, weekly group meetings, journal clubs and to external partners wherever involved.
